I'm running 5.5 on few hosts (including one with 30GB database) and there is 
no problem.

Remember that if you want to upgrade without dump & restore you have to 
upgrade from series to next series for example:

4.1 -> 5.0; run mysql_upgrade script
5.0 -> 5.1; run mysql_upgrade script
5.1 -> 5.5; run mysql_upgrade script

You cannot upgrade directly from 5.0 to 5.5 for example.

5.5 will land in main this week.
